PEPTIDES FOR ANTI-AGING COSMETICS IN CLINICAL DEVELOPMENT

Peptides have emerged as popular active ingredients in cosmetics in order to help maintain a healthy and youthful appearance. Some peptides affect various cellular and molecular processes within the skin through modulation of collagen, elastin and melanin. In particular, the ability of some synthetic peptides to increase or maintain production of dermal extracellular matrix proteins such as collagen is of interest for anti-aging applications (1). Furthermore, researchers have shown that several peptides have the ability to promote wound healing, cell migration, and angiogenesis. Many peptides are active ingredients in marketed cosmetic products but often there is little in vivo efficacy data known. In addition, cosmetic peptides are often tested as part of a mixture or as a cosmetic formulation limiting knowledge pertaining to the effect of an individual peptide (2). Several examples of cosmetic peptides that have undergone clinical studies to confirm efficacy on human skin are shown in Table 1.

Collagen Stimulators

Progeline (Trifluoroacetyl-tripeptide-2) was studied in two in vivo studies. In one study, Progeline was shown to reduce the volume of jawlines after 4 weeks and 8 weeks of treatment. In the second study, the tripeptide improved cutaneous firmness and visco-elasticity. The anti-wrinkle and anti-sagging effects of Progeline treatment were also confirmed by facial macrophotography (3).

Decorinyl® (Tripeptide-10 Citrulline) mimics decorin, a proteoglycan that binds to collagen fibers. This tetrapeptide regulates collagen fibrillogenesis and the growth of collagen fibers making them thinner and uniform. Results from a trial comparing a Decorinyl containing cream to placebo showed an increase of skin suppleness by 54% (4).

Matrixyl® 3000 is an anti-wrinkle peptide mixture containing pal-GHK tripeptide and pal-GQPR tetrapeptide. A clinical study comparing a Matrixyl 3000 containing cream to a placebo cream confirmed anti-wrinkle efficacy and other benefits such as reduction of wrinkle depth, volume and decrease in the area occupied by deep wrinkles (5).

 Carrier Peptides

Copper Tripeptide (GHK-Cu) is widely used in skin and hair products. This peptide has multiple actions such as stimulating blood vessel and nerve growth. It also increases collagen, elastin and glycosaminoglycan synthesis. The cosmetic benefits of Copper Tripeptide have been confirmed in many clinical studies. In one study, a Copper Tripeptide eye cream applied to photo-damaged eye skin was shown to reduce lines and wrinkles, improve appearance, and increase skin density and thickness (6).

 Neurotransmitter Inhibitor Peptides

Argireline® (Acetylhexapeptide-3) is a well-known botox-like cosmetic peptide. This peptide is reported to inhibit the release of neurotransmitters and reduce the degree of facial wrinkles. Argireline is the N-terminal end of the synaptosomal nerve-associated protein 25 (SNAP-25) and through competition with the natural protein, the peptide destabilizes the SNARE complex which inhibits release of neurotransmitters and attenuates muscle contraction. In a study of Argireline versus placebo, the anti-wrinkle efficacy in the Argireline treated group was measured at 48.9%, compared with 0% in the placebo group (7).

Leuphasyl® (Pentapeptide-18) is another peptide that has botox-like effects. This peptide mimics enkephalin action, inhibits neuronal activity, and inhibits catecholamine release. In one in vivo study, Leuphasyl was found to have efficacy at a minimum of 2% concentration. In addition, study results showed that the peptide is free from side effects and attenuates wrinkles while also preserving facial expressivity (8).

SYN®-AKE (Tripeptide-3) is a small synthetic peptide that mimics the effect of a peptide found in the venom of the viper Tropidolaemus wagleri. This tripeptide is a reversible antagonist of the muscle-type nicotinic acetylcholine receptor (mnAChR). As a cosmetic active ingredient, it allows the face to relax and smooths out wrinkles. Results from a clinical study showed that SYN-AKE reduced the appearance of wrinkles after 28 days by up to 52% (9).

 Conclusion

Peptides are a growing area of interest in the field of cosmetics for anti-aging and other personal care applications. While some cosmetic peptides are widely studied and used as active ingredients in the cosmetic industry, clinical studies proving the efficacy of cosmetic peptides are not always published in scientific journals. What is your official job title at Bachem?

My official job title is Market Analyst.

 

How long have you been with Bachem? Where did you work before Bachem?

I am with Bachem since September 2019. Before joining Bachem, I worked in the Marketing & Sales department as a Business Analyst for a large chemical company in Switzerland.

 

Briefly, what do you do at Bachem?

I focus on Business Intelligence and Competitive Intelligence. I evaluate data, as well as competitor and industry data, to discover ways to improve Bachem’s market position.

 

What is your academic background/degrees or training?

I hold both a Bachelor and a Master degree in business management.

 

What do you like to do outside of work?

I enjoy spending time with family and friends. It is a great way to socialize and to stay motivated by encouraging each other.

 

How is the Marketing & Sales team partnering with their customers?

Steady communication with customers helps to gain better understanding of their requirements, and overall make customers feel valued and important.
